胰岛素样生长因子-Ⅰ调节胚胎着床的机理研究

摘　要：进一步研究IGF-1对整合素介导的信号转导通路的调节作用,探讨胰岛素样生长因子-Ⅰ调节胚胎着床的作用机理.方法:在参照文献[1]的基础上,采用免疫细胞化学SP法检测细胞中信号分子磷酸化粘着斑激酶的定性表达,采用免疫印迹(Western Blot)方法检测磷酸化粘着斑激酶的定量表达.结果:免疫组化染色显示:滋养层细胞加入IGF-Ⅰ培养后,细胞中信号分子磷酸化粘着斑激酶呈阳性表达.免疫印迹结果显示:IGF-Ⅰ处理后,滋养层细胞中磷酸化粘着斑激酶的表达量随浓度的增加和时间的延长而增多.抗整合素β3抗体则阻止磷酸化粘着斑激酶的表达.结论:IGF-Ⅰ通过激活磷酸化粘着斑激酶进而激活整合素--磷酸化粘着斑激酶的信号传导通路,促进滋养层细胞黏附到纤维粘连蛋白.山西煤炭中心医院生殖与遗传医学中心弓爱东
